Techniques for measuring the circular dichroism in vibrational transitions (VCD) using Fourier transform infrared (FT-IR) spectroscopy have been developed recently. While measuring VCD on FT-IR spectrometers, one generally encounters problems associated with polarization artifacts that arise from the birefringence in the optical path and detector window. Until now, these problems were overcome by recording VCD either as one half of the difference between the VCD spectra of the enantiomers or as the difference between the VCD spectra of one enantiomer and the racemic mixture. When neither the second enantiomer nor the racemic mixture is available, as for most biological compounds, this procedure is inapplicable.
