What determines the success of isomerization of the Photoactive Yellow Protein chromophore? – A picosecond pump-probe study in the midIR

Abstract

We have measured the mid-infrared absorption-difference spectra of PYP photocycle intermediates representing successful and unsuccessful attempts to enter the signaling state. It appears that breaking of the chromophores hydrogen bond with Cys69 is decisive.
