Near-field single molecule spectroscopy

Abstract

Recent advances in near-field scanning optical microscope (NSOM)1 make it possible to image single molecule emission with nanometric resolution2 and extend the single molecule spectroscopy work done at cryogenic temperature3 to the room temperature regime. With NSOM, molecular fluorescence can be spectrally4 and temporally resolved,5-6 allowing detailed studies of molecular interaction and dynamics on a single molecule basis with a spatial resolution beyond the diffraction limit. Experiments probing the dynamics of single molecules on both the 10-2 to 103 sec time scale and the picosecond to nanosecond time scale have ben demonstrated.
