Abstract

Optically trapped beads with diameters of 100 nm to 500 nm are scanned across surface structures. The elongation of the probe from the trap center is measured interferometrically so that the height profile of the sample can be recovered. The Brownian motion of the probe helps to find the local force field and viscosity.
