Localizing and Tracking Single Emitters in Three Dimensions Using a Double Helix Point Spread Function

Abstract

Single fluorescent emitters can be localized and tracked in 3D with less than 1000 detected photons by using a DH PSF. The tracking of quantum dots both in solution and inside a living cell is demonstrated.
