Capillary substrate of brain tissue oxygenation changes with age

Abstract

This work aimed to investigate the impact of vascular changes with age on brain tissue oxygenation. In a first set of experiments, we measured capillary RBC dynamics in sensory-motor cortex of young and aged mice under isoflurane-anesthetized conditions and reproduced previous results found in rats. In a separate cohort we then measured the impact of these changes on tissue oxygenation using two-photon lifetime imaging of the phosphorescent probe PtP-C343. Our results show decreased oxygen delivery and brain tissue oxygenation with age suggesting a role for oxygen delivery in cognitive decline.
