Abstract
We introduce a photonic crystal enhanced microscope (PCEM) (Figure 1a) as an imaging approach that combines enhanced fluorescence (EF) and label-free (LF) imaging modalities for assays performed upon a photonic crystal (PC) surface (Figure 1b). The instrument utilizes a collimated laser illumination source and tuning of the incidence angle to enable highly efficient coupling to PC optical resonances. The system has been used for label-free imaging of DNA and protein capture spots for microarray quality control, while amplifying signals from fluorescent-tagged hybridized biomolecules by >500× compared to performing the same assay on an ordinary glass surface. The same system has also been used for imaging cell surface attachment to the PC, and measuring how drugs modulate cell attachment.
