Abstract

The applicability of total-light algorithm has been tested with tissue phantoms. It was experimentally confirmed that this algorithm can construct the background image in the absence of the fluorescence target from temporal profiles of the excitation and the fluorescence in the presence of the target. Then, the absorption image of the fluorescent target could be successfully obtained. This method can be useful in the quantification of the fluorophores in inhomogeneous material, such as whole tissues.
